研究方向
真核复杂类型细胞如干细胞/免疫细胞等基因转录后调控的机制研究正越来越受到研究者的关注。相比转录水平(组蛋白修饰,DNA修饰,转录因子等),转录后调控(RNA加工,稳定性调控等)对于细胞来说,它消耗能量更少,更便于细胞精细调控,满足复杂细胞应对发育分化/抗炎应急等外来信号的需求。本研究组近期围绕mRNA APA(alternative polyadenylation)的机制和生物学功能意义展开以下科学问题的探索。
1. 非编码RNA调控mRNA3'末端加工和APA的机制
2. mRNA3'末端加工因子或APA调控因子是否参与干细胞命运决定过程
3. mRNA3'末端加工因子或APA调控因子是否参与某些病理过程
学术成果
主要学术研究成果与贡献
(1)首次发现U4/U6/U2 snRNP具有类似U1抑制pre-mRNA PCPA (premature cleavage and polyadenylation)和PTT(premature transcription termination)的功能,从而在共转录加工的分子机制领域提出普适性模型:pre-mRNA splicing在转录组规模可防止PCPA/PTT,从而正调控transcription。相关结果以最后通讯作者发表在国际权威期刊《PNAS》2024和《Cell Reports》2025上。
(2)2023年在生化领域权威期刊《JBC》上以最后通讯作者发表U1 snRNP telescripting的分子机制论文,为U1抑制pre-mRNA PCPA与PTT带来最新的见解。
(3)2017年在国际知名刊物《NAR》以最后通讯作者发表snoRNA概念与功能突破性进展研究,Article被评为NAR Breakthrough Article.首次揭示snoRNA可能在真核生物mRNA3'末端加工中发挥功能。
